Ok., I'm going to be biased again. I've never used Glade. I do
program a bit of GTK+, but again only in Emacs. Usually one of my
friends does the packing of widgets. He uses Glade for that. But
once the code gen is done, he copies the code into the project and
uses good ol` Emacs to continue with it.
I find Emacs more comfortable than using glade. I'm used to
writing my own autotools based build system., so I find the build
system given by Glade as cumbersome..
